RESUMO

Electrochemically in-situ generation of oxygen and caustic soda is promising for sulfide management while suffers from scaling, poor inactivating capacity, hydrogen release and ammonia escape. In this study, the four-compartment electrochemical cell efficiently captured oxygen molecules from the air chamber to produce HO2- without generating toxic by-products. Meanwhile, the catalyst layer surface of PTFE/CB-GDE maintained a relatively balanced gas-liquid micro-environment, enabling the formation of enduring solid-liquid-gas interfaces for efficient HO2- electrosynthesis. A dramatic increase in HO2- generation rate from 453.3 mg L-1 h-1 to 575.4 mg L-1 h-1 was attained by advancement in operation parameters design (flow channels, electrolyte types, flow rates and circulation types). Stability testing resulted in the HO2- generation rate over 15 g L-1 and the current efficiency (CE) exceeding 85%, indicating a robust stable operational capacity. Furthermore, after 120 mg L-1 HO2- treatment, an increase of 11.1% in necrotic and apoptotic cells in the sewer biofilm was observed, higher than that achieved with the addition of NaOH, H2O2 method. The in-situ electrosynthesis strategy for HO2- represents a significance toward the practical implementation of sulfide abatement in sewers, holding the potential to treat various sulfide-containing wastewater.

The productivity and efficiency of two-chamber microbial electrolysis cell and anaerobic digestion integrated system (MEC-AD) were promoted by a complex of anaerobic granular sludge and iron oxides (Fe-AnGS) as inoculum. Results showed that MEC-AD with Fe-AnGS achieved biogas upgrading with a 23%-29% increase in the energy recovery rate of external circuit current and a 26%-31% decrease in volatile fatty acids. The energy recovery rate of MEC-AD remained at 52%-57%, indicating a stable operation performance. The selectively enriched methanogens and electroactive bacteria resulted in dominant hydrogenotrophic and acetoclastic methanogenesis in the cathode and anode chambers. Mechanistic analysis revealed that MEC-AD with Fe-AnGS led to specifically upregulated enzymes related to energy metabolism and electron transfer. Fe-AnGS as inoculum could improve the long-term operation performance of MEC-AD. Consequently, this study provides an efficient strategy for biogas upgrading in MEC-AD.

New chiral ligands could be obtained by introducing proline moieties and imidazoline moieties to binaphthyl skeletons. The chiral ligands exhibited balanced rigidity and flexibility which could allow the change of the conformations during the reactions on one hand, and could provide sufficient asymmetric induction on the other. The proline moiety could act as a linker connecting the binaphthyl skeletons and the imidazoline moieties as well as a coordinating group for the central metal, and the electronic and steric properties of the imidazoline groups could be carefully fine-tuned by the use of different substituents. In the presence of Cu(II) catalyst bearing such chiral ligands, aza-Friedel-Crafts reaction of 1-naphthols and electron-rich phenols with isatin-derived ketimines provided the desired products with good to excellent yields and up to 99 % ee. The reactions showed good scalability, and excellent ee could still be obtained when the reaction was carried out in gram-scale.

Microbial electrolysis cells (MECs) are increasingly recognized as a promising technology for converting CO2 to CH4, offering the dual benefits of energy recovery from organic wastewater and CO2 emission reduction. A critical aspect of this technology is the enhancement of the electron-accepting capacity of the methanogenic biocathode to improve CH4 production efficiency. This study demonstrates that adjusting the cathode resistivity is an effective way to control the electric field intensity, thereby enhancing the electron accepting capacity and CH4 production. By maintaining the electric field intensity within approximately 8.50-10.83 mV·cm-1, the CH4 yield was observed to increase by up to two-fold. The improvement in CH4 production under optimized electric field conditions was attributed to the enhancement of the direct accepting capacity of the biocathode. This enhancement was primarily due to an increase in the relative abundance of Methanosaeta by approximately 10 % and an up to 83.78 % rise in the electron-accepting capacity of the extracellular polymeric substance. These insights offer a new perspective on the operation of methanogenic biocathodes and propose a novel biocathode construction methodology based on these findings, thus contributing to the enhancement of MEC efficiency.

Sulfur-containing substances in sewers frequently incur unpleasant odors, corrosion-related economic loss, and potential human health concerns. These observations are principally attributed to microbial reactions, particularly the involvement of sulfate-reducing bacteria (SRB) in sulfur reduction process. As a multivalent element, sulfur engages in complex bioreactions in both aerobic and anaerobic environments. Organic sulfides are also present in sewage, and these compounds possess the potential to undergo transformation and volatilization. In this paper, a comprehensive review was conducted on the present status regarding sulfur transformation, transportation, and remediation in sewers, including both inorganic and organic sulfur components. The review extensively addressed reactions occurring in the liquid and gas phase, as well as examined detection methods for various types of sulfur compounds and factors affecting sulfur transformation. Current remediation measures based on corresponding mechanisms were presented. Additionally, the impacts of measures implemented in sewers on the subsequent wastewater treatment plants were also discussed, aiming to attain better management of the entire wastewater system. Finally, challenges and prospects related to the issue of sulfur-containing substances in sewers were proposed to facilitate improved management and development of the urban water system.

Rice blast, caused by Magnaporthe oryzae, significantly impacts grain yield, necessitating the identification of broad-spectrum resistance genes and their functional mechanisms for disease-resistant crop breeding. Here, we report that rice with knockdown OsHDAC1 gene expression displays enhanced broad-spectrum blast resistance without effects on plant height and tiller numbers compared to wild-type rice, while rice overexpressing OsHDAC1 is more susceptible to M. oryzae. We identify a novel blast resistance transcription factor, OsGRAS30, which genetically acts upstream of OsHDAC1 and interacts with OsHDAC1 to suppress its enzymatic activity. This inhibition increases the histone H3K27ac level, thereby boosting broad-spectrum blast resistance. Integrating genome-wide mapping of OsHDAC1 and H3K27ac targets with RNA sequencing analysis unveils how OsHDAC1 mediates the expression of OsSSI2, OsF3H, OsRLR1 and OsRGA5 to regulate blast resistance. Our findings reveal that the OsGRAS30-OsHDAC1 module is critical to rice blast control. Therefore, targeting either OsHDAC1 or OsGRAS30 offers a promising approach for enhancing crop blast resistance.

In response to the problem of asynchronous fermentation between lignocellulose and perishable materials in compost, the combined technology of low-temperature hydrochar and compost has been studied. Hydrochar was prepared through low-temperature hydrothermal reactions and applied to aerobic fermentation. The response relationship between lignocellulose content, electron transfer capability, and microbes was explored. The results showed that a pore structure with oxygen-containing functional groups was formed in hydrochar, promoting electron transfer during composting. With the rapid increase in composting temperature, the lignocellulose content decreased by 64.36 mg/g. Oceanobacillus, Cerasibacillus, Marinimicrobium, and Gracilibacillus promoted the degradation of lignocellulose and the carbon/nitrogen cycle during aerobic fermentation, and there was a significant response relationship between electron transfer capability and functional microbes. The combined application of hydrochar and aerobic fermentation accelerated the degradation of lignocellulose. This study provides technical support for the treatment of heterogeneous organic waste.

OBJECTIVE: Neuromyelitis optica spectrum disorders (NMOSD) is often misdiagnosed or delayed because of the complex and diverse clinical manifestations, especially the atypical initial presentation. Hyponatremia can be an infrequently isolated initial presentation of NMOSD and is associated with hypothalamus involvement. Awareness of this mechanism will help clinicians to identify NMOSD early, treat it in time and improve the prognosis. METHODS: We describe a 36-year-old woman who developed repeated hyponatremia and then experienced diplopia. Serum AQP4, MOG, MBP and GFAP antibody were detected, and NMOSD was finally diagnosed. RESULTS: She responded well to high-dose glucocorticoids. Sequential treatment with mycophenolate mofetil (MMF) was prescribed. Two-month follow-up revealed full recovery. So far, after 10 months, the patient still has no recurrence. CONCLUSION: For young patients, repeated hyponatremia, with or without slight fever, and no evidence of obvious infection, brain magnetic resonance imaging (MRI) and serum AQP4/MOG antibody detection may be useful to determine whether there is a possibility of NMOSD.

BACKGROUND: The aleurone layer is a part of many plant seeds, and during seed germination, aleurone cells undergo PCD, which is promoted by GA from the embryo. However, the numerous components of the GA signaling pathway that mediate PCD of the aleurone layers remain to be identified. Few genes and transcriptomes have been studied thus far in aleurone layers to improve our understanding of how PCD occurs and how the regulatory mechanism functions during PCD. Our previous studies have shown that histone deacetylases (HDACs) are required in GA-induced PCD of aleurone layer. To further explore the molecular mechanisms by which epigenetic modifications regulate aleurone PCD, we performed a global comparative transcriptome analysis of embryoless aleurones treated with GA or histone acetylase (HAT) inhibitors. RESULTS: In this study, a total of 7,919 differentially expressed genes (DEGs) were analyzed, 2,554 DEGs of which were found to be common under two treatments. These identified DEGs were involved in various biological processes, including DNA methylation, lipid metabolism and ROS signaling. Further investigations revealed that inhibition of DNA methyltransferases prevented aleurone PCD, suggesting that active DNA methylation plays a role in regulating aleurone PCD. GA or HAT inhibitor induced lipoxygenase gene expression, leading to lipid degradation, but this process was not affected by DNA methylation. However, DNA methylation inhibitor could regulate ROS-related gene expression and inhibit GA-induced production of hydrogen peroxide (H2O2). CONCLUSION: Overall, linking of lipoxygenase, DNA methylation, and H2O2 may indicate that GA-induced higher HDAC activity in aleurones causes breakdown of lipids via regulating lipoxygenase gene expression, and increased DNA methylation positively mediates H2O2 production; thus, DNA methylation and lipid metabolism pathways may represent an important and complex signaling network in maize aleurone PCD.

RESUMO

The copper(II)-catalyzed enantioselective aza-Friedel-Crafts reaction of indoles with isatin-derived N-Boc-ketimines was developed by using tunable chiral O-N-N tridentate ligands derived from BINOL and proline. In general, the reaction afforded chiral 3-indolyl-3-aminooxindoles under mild conditions in high yields (83-97%) with excellent ee (69-99%).

Iron-based peracetic acid (PAA) advanced oxidation process (AOP) is widely used in water purification because of its high efficiency and low toxicity. In this study, for the first time, ferrous iron (Fe2+) and PAA were dosed jointly into the rising main sewer reactor, to verify the feasibility of sulfide and methane control as well as investigate the comprehensive mechanism of Fe2+/PAA on sewer biofilm. Results demonstrated the superior biocidal effect of Fe2+/PAA dosing than that of PAA alone. Intermittent Fe2+/PAA dosing showed that the average inhibitory rate of sulfide production rate (SPR) and methane production rate (MPR) was 52.0% and 29.9%, respectively, at a Fe2+/PAA molar ratio of 1:1 and PAA concentration of 3 mmol/L (i.e., the mass-based concentrations of Fe2+ and PAA were 6.79 mg-Fe/L and 228 mg/L, respectively). Beside, sewer biofilm was found to be resistant to PAA during repeated dosing events. However, resistance could be alleviated by introducing sulfide in situ in the Fe2+/PAA process, and SPR and MPR were further reduced to 27.39% and 67.32% of the control, respectively. LIVE/DEAD Staining showed that Fe2+/PAA exhibited a strong destructive effect on microbial cells, with the proportion of viable cells being 26.34%. Electron paramagnetic resonance (EPR) and free radical quenching results indicated that the inhibitory order was R-O• > •OH > Fe(IV), which led to the disruption of cellular integrity (i.e., 17.24% increase in LDH) and intracellular enzyme system (i.e., cellular metabolic disorders). Microbial analysis revealed that long-term Fe2+/PAA dosing decreased the sulfate-reducing bacteria (SRB) abundance, and the dominant genus of methanogenic archaea (MA) shifted from Methanofastidiosum, Methanobacterium to Methanosaeta. The cost of Fe2+/PAA dosing on methane and sulfide control in rising main sewers was $1.81/kg-S, economically and environmental-friendly attractive for practical applications.

Ferric chloride (FeCl3) served as a solid acid has attracted attention recently. However, the feasibility of FeCl3 combined with nitrite for free nitrous acid (FNA) generation in controlling sulfide and methane as well as the triggering mechanisms in the complex syntrophic consortium (i.e., sewer biofilm) remain largely unknown. This work disclosed FeCl3 as an alternative acid source could obtain comparable sulfide and methane mitigations at a low FNA dose (i.e., 0.26 mg N/L), compared to that of HCl acid source. Whereas, a faster recovery rate of sulfide production was observed using FeCl3 under a higher FNA dose (i.e., 0.81 mg N/L) despite the methane control still being comparable. The toxicological mechanisms revealed FNA reacted with proteins amide Ⅰ in extracellular polymeric substances and destroyed protein hydrogen bond. Enzymatic and genic analysis unveiled the overall suppression of hydrolysis, acidogenesis, acetogenesis, sulfidogenesis and methanogenesis steps due to the inactivation of viable cells by reactive nitrogen species. Economic and environmental assessments demonstrated that the ferric-based FNA strategy reduced chemical costs and N2O emission (ca. 26.5% decrease) compared to the traditional HCl-based FNA method. This work broadens the application of iron salt-based technology in urban water system, together with understanding the biological mechanisms of FNA-based technology.

Microplastics (MPs) can accumulate in animal organs. Numerous studies have linked MPs with immune system. However, the impact of MPs on immune response remains unclear. This study examined the innate immune response of mice exposed to 5 µm MPs. In the lipopolysaccharide challenge, mice treated with MPs exhibited lower levels of serum immune factors and activated immune cells. MPs disrupted immune-related receptors and cause dysfunction in cell signal transduction within the liver and spleen. Proteomic analysis revealed that MPs impede the activation of serum immune-related signals. In addition, the tissue section imaging exhibited a significant enrichment of MPs in the extracellular matrix (ECM), consistent with the ECM dysfunction and immune receptor suppression. Therefore, our data suggest excessive MPs accumulation in ECM inhibits cell signaling pathways, thereby suppressing the activation of immune responses. We propose the biotoxicity of MPs is partly through the MP disruption of ECM (MPDEM).

Microplastic (MP) pollution has become a global environmental problem, with particular concerns for its harmful effects on human health. Several studies have demonstrated that MP can penetrate animals and humans resulting in tissue dysfunction, but their influences on metabolism remain poorly understood. In this study, we investigated the impact of MP exposure on metabolism and the results showed that different treatment doses produce a bidirectional modulatory effects on mice. When exposed to high concentrations of MP, mice lost significant weight, while those in the lowest concentration treatment group showed little change, but those treated at relatively low concentrations became overweight. There was excessive lipid accumulation in these heavier mice, with a better appetite and lower activity level. Transcriptome sequencing revealed that MPs increased fatty acid synthesis in the liver. In addition, the gut microbiota composition of the MPs-induced obese mice was remodeled, which would enhance the nutrient absorption capacity of the intestine. Our results uncovered an MP dose-dependent lipid metabolism in mice and a non-unidirectional model of the physiological responses to different MP concentrations was proposed. These results provided new insights into the seemingly contradictory effects of MP on metabolism in the previous study.

Using binaphthyl-proline-based chiral ligands, Zn(II)-catalysed addition of alcohols and tert-butyl hydroperoxide to isatin-derived N-Boc ketimines provided the isatin-derived C3 N,O-aminals in up to 99% yield and up to 99% ee. The reactions could be carried out under mild conditions and a gram-scale reaction could be realized without the loss of the yield and enantioselectivity.

Although the biocidal effect of calcium peroxide (CaO2) has attracted increasing attention in wastewater and sludge management, its potential in the reduction of sulfide and methane from sewer is not tapped. This study aims to fill this gap through the long-term operated sewer reactors. Results showed one-time dose of 0.2% (w/v) CaO2 with 12-h exposure decreased the average sulfide and methane production by 80% during one week. The electron paramagnetic resonance and free radical quenching tests indicated free radicals from CaO2 decomposing posed a major contribution on sewer biofilms (•OH>•O2->alkali). Mechanistic analysis revealed extracellular polymeric matrix breakdown (e.g., protein secondary structure) and cell membrane damage were caused by the increased lipid peroxidation of cells and exacerbated intracellular reactive oxygen species under CaO2 stress. Moreover, the intracellular metabolic pathways, such as electrons provision and transfer, as well as pivotal enzymatic activities (e.g., APS reductase, sulfite reductase and coenzymes F420) were significantly impaired. RT-qPCR analysis unveiled the absolute abundances of dsrA and mcrA were decreased by 7.53-40.37% and 67.00-74.85%, respectively. Although this study broadens the application scope of CaO2 and provides in-depth understanding of advanced oxidation-based technology in sewer management, the pipe scale risk due to the release of calcium ions warrants further investigation.

New Cu(I) catalysts are effective in enantioselective Friedel-Crafts alkylation of a variety of indoles with different ß,γ-unsaturated α-ketoesters. A control study shows that such a catalyst system is less sensitive to air, and the reactions can be carried out without special cares such as glovebox operation or moisture/oxygen-free conditions. Preliminary computation results suggest that there exists π-π stacking between the substrate and the catalyst, and such an interaction seems to play a role in stabilizing the reaction intermediate and enhancing the stereoselectivity of the reactions. The desired products can be obtained in up to 98% yield at 99% enantiomeric excess. The same high enantioselectivity can be observed when the reaction is carried in a gram scale, indicating a good scalability of the catalyst system in enantioselective Friedel-Crafts alkylation of different indoles with ß,γ-unsaturated α-ketoesters.

Abelmoschus manihot L. (HSK) is a rare and endangered species in the wild that grows on the cliffs of deep mountains. As a natural plant, the chemical composition of HSK is relatively complex, which mainly includes flavonoids, organic acids, polysaccharides, and various trace elements with good effects of clearing away heat, anti-inflammatory, analgesic, and calming nerves, and inhibiting tumor cells. In this experiment, different developmental stages of HSK flowers were used for optimization of the flavonoid extraction and determining method. The antioxidant activities, flavonoid accumulation pattern, and synthesis regulatory network were analyzed using biochemistry, RNA-seq, and UPLC-MS/MS. The total content of flavonoids, vitexin rhamnoside, hyperoside, and rutin in HSK flowers at T3 stage (flower wilting) was significantly higher than in T2 (full flowering) and T1 (bud) stages. Compared with T1 and T2, the antioxidant capacity of the T3 flower alcohol extract was also the strongest, including the total reducing ability, DPPH clearance, OH clearance, O2- clearance, and total antioxidant capacity. A total of 156 flavonoids and 47,179 unigenes were detected by UPLC-MS/MS and RNA-Seq, respectively. The candidate genes and key metabolites involved in flavonoid biosynthesis were identified and the regulatory networks were also analyzed in this study. qRT-PCR test further proved that the gene expression level was consistent with the results of RNA sequence data. The relationship between the gene expression and flavonoid accumulation network provides a theoretical basis for the mining and regulation of functional genes related to the flavonoid biosynthesis and metabolism in Abelmoschus manihot L.

The CONSTITUTIVE PHOTOMORPHOGENIC9 (COP9) signalosome (CSN) is a multi-functional protein complex, which is involved in plant growth and abiotic stress response. However, the evolution and function of the CSN genes in land plants are still largely unclear. Here, we have identified 124 CSN genes and constructed phylogenetic trees of these CSN proteins to elucidate their feature and evolution in twelve land plants. Analysis of gene structure, protein property and protein motif composition shows the evolutional conservation and variation of the CSN proteins in land plants. These CSN genes might evolve through whole genome duplication (WGD)/segmental duplication (SD) and tandem duplication (TD). Analysis of promoter cis-elements shows that the CSN genes are implicated in diverse biological processes and different signaling pathways. RT-qPCR indicates that the transcript abundance of the OsCSN genes is up-regulated or down-regulated in response to abiotic stress and treatment with various hormones in rice. These results provide new insights into the CSN gene evolution and its possible function in land plants.

The potential of periodate (PI) in sludge anaerobic digestion is not tapped, although it has recently attracted great research interest in organic contaminants removal and pathogens inactivation in wastewater treatment. This is the first work to demonstrate significant improvement in methane generation from waste activated sludge (WAS) with PI pretreatment and to provide underlying mechanisms. Biochemical methane potential tests indicated that methane yield enhanced from 100.2 to 146.3 L per kg VS (VS, volatile solids) with PI dosages from 0 to 100 mg per g TS (TS, total solids). Electron spin resonance showed PI could be activated without extra activator addition, which might be attributed to the native transition metals (e.g., Fe2+) in WAS, thereby generating hydroxyl radical (•OH), superoxide radicals (•O2 -), and singlet oxygen (1O2). Further scavenging tests demonstrated all of them synergistically promoted WAS disintegration, and their contributions were in the order of •O2 - > •OH > 1O2, leading to the release of substantial biodegradable substances (i.e., proteins and polysaccharides) into the liquid phase for subsequent biotransformation. Moreover, fluorescence and ultraviolet spectroscopy analyses indicated the recalcitrant organics (especially lignocellulose and humus) could be degraded by reducing their aromaticity under oxidative stress of PI, thus readily for methanogenesis. Microbial community analysis revealed some microorganisms participating in hydrolysis, acidogenesis, and acetoclastic methanogenesis were enriched after PI pretreatment. The improved key enzyme activities and up-regulated metabolic pathways further provided direct evidence for enhanced methane production. This research was expected to broaden the application scope of PI and provide more diverse pretreatment choices for energy recovery through anaerobic digestion.
